DescriptionDiabetic foot ulcers (DFUs) represent a major complication of diabetes mellitus with limited therapeutic options and high clinical burden. Endothelial dysfunction and impaired wound healing are hallmark features of DFU pathogenesis, yet the underlying molecular mechanisms remain incompletely understood. In this study, we performed integrative proteomic and interactomic analyses to investigate the regulatory network governing endothelial mitophagy in DFU. Using immunoprecipitation coupled with mass spectrometry (IP-MS) and quantitative proteomics, we identified claudin 8 (CLDN8) as a critical regulator of endothelial function and wound repair. Our multi-omics dataset reveals that CLDN8 activates MAPK/ERK/c-MYC signaling to transcriptionally upregulate USP28, which subsequently deubiquitinates and stabilizes the mitophagy receptor FUNDC1. This signaling cascade promotes selective mitochondrial autophagy, preserves endothelial metabolic homeostasis, and accelerates wound closure. In vivo validation in diabetic rat models confirmed the therapeutic potential of CLDN8 restoration. The raw mass spectrometry data and processed results have been deposited in this project, including IP-MS datasets for CLDN8-interacting proteins and global proteomic profiling of endothelial cells under diabetic conditions. These datasets provide a comprehensive resource for understanding the molecular basis of endothelial mitophagy and may facilitate the development of novel therapeutic strategies for DFU intervention.
