TitleMonocyte phosphoproteomics for mechanosensory shear stress
DescriptionCalcific aortic valve disease (CAVD) exposes circulating monocytes to pathological haemodynamic forces before tissue infiltration, yet the molecular mechanisms by which these forces regulate inflammatory signalling remain poorly understood. Using our established microfluidic model of aortic stenosis, human monocytes were exposed to physiological or pathological shear stress. Quantitative proteomic and phosphoproteomic analyses identified mechanosensitive pathways activated by pathological shear stress, which were validated in primary human monocytes and paired monocytes isolated from patients with CAVD before and after transcatheter aortic valve implantation (TAVI). Perseus (v2.0.7.0) used for global proteomics and phosphoprotoemics data analysis.
