| Title | A CTC cluster group with single-cell metabolic signature and metastatic mechanism transition favors TNBC prognosis prediction |
| Description | Triple-negative breast cancer (TNBC) patients easily experience recurrence and metastasis after treatment. Circulating tumor cell (CTC) counts may not be the most powerful tool to predict a favorable prognosis. However, simple classification of CTCs into clusters and single cells due to their apparent heterogeneity is insufficient. Underlying factors such as single-cell metabolic signature, may contribute more directly to the metastatic capability of CTCs. Herein, we first identified signature metabolites between TNBC single cell and cell cluster models. Subsequently, we used a homebuilt single-cell quantitative mass spectrometry platform for quantifying signature metabolites of each CTC in TNBC patients. CTCs were further classified into three groups based on the abundance of the metabolic signature at the single-cell level. After construction of a prognosis prediction model based on the CTC groups and clinical variables, a specific CTC cluster group emerged as a significant contributor to TNBC prognosis. Herein, particular effort was made to explore the impact of hypoxia on this metabolic classification both in vitro and in vivo, which further elucidated the molecular mechanism transition underlying metabolic reprogramming during cancer metastasis and led to the identification of potential therapeutic target in TNBC. |
