Title	IP-MS Detection of His-cTGF-Binding Membrane Proteins
Description	This project aims to detect membrane proteins that bind to CTGF in cells. The specific procedure is as follows: First, 18 10-cm HCC1806 culture dishes were starved overnight in serum-free medium, then treated with 100 ng/ml His-CTGF at 4°C for 2 hours (two groups: -CTGF and +CTGF; each group required 3 10-cm dishes, with three replicates). Following this, membrane proteins were extracted using a membrane protein extraction kit. Next, 200 μL of Biyuntian RIPA lysis buffer (containing PMSF) was added to each tube, and binding was performed overnight at 4°C using His-tag-conjugated magnetic beads. Elution was performed in 10-minute intervals, with each tube eluted using 30 μL of 2× SDS. The samples were then subjected to Western blot analysis, followed by gel cutting and mass spectrometry identification (IP-MS).
