TitleA Multiplex MRM Quantitative Method for Forensic Body Fluid Identification
DescriptionThis study was performed based on the Shimadzu LCMS-8050 high-performance liquid chromatography coupled with triple quadrupole tandem mass spectrometry (HPLC-MS/MS) platform. A targeted multiplex peptide detection method utilizing high-flow-rate reversed-phase chromatography was established. Synthetic peptides were adopted to optimize the analytical workflow, and stable isotope-labeled internal standard peptides were introduced to achieve reliable absolute quantification of 21 body fluid-specific peptides. The established peptide absolute quantification strategy was further applied to forensic identification of body fluid and tissue origins. In addition, three types of control peptides were incorporated into the assay. Bovine-derived CYCS protein was used as a negative quality control marker. Matrix samples spiked with bovine serum albumin (BSA) were repeatedly injected to assess the method’s accuracy and precision. Meanwhile, matrix-matched samples prepared from peripheral blood, menstrual blood, vaginal secretions, semen and saliva were analyzed to evaluate matrix effects.
