TitleVisual exoproteomics of Clostridium thermocellum during anaerobic biomass-degradation identifies functional spirosomes
DescriptionThis project utilizes a visual proteomics approach to characterize the exoproteome of the anaerobic, cellulose-degrading bacterium Clostridium thermocellum during biomass degradation. By integrating mass spectrometry with high-resolution cryo-electron microscopy (cryo-EM), we interrogated the composition of large exoproteome assemblies and extracellular machinery. Mass spectrometry analysis confirmed a broad exoproteome composition, notably identifying key cellulose-degrading components such as CelA and CipA. Further proteomic investigation of high-abundance protein filaments and vesicular structures, combined with de novo modeling, identified the presence of functional aldehyde-alcohol dehydrogenase (AdhE) spirosomes. This dataset contains the mass spectrometry evidence used to support protein identification and structural characterization of these extracellular complexes, providing insight into the functional exoproteome landscape of C. thermocellum.
