| Title | The Effect of Surface Coating on Microbial Community Diversity and Barnacle Fouling at Varying Salinities |
| Description | Samples were adjusted to 1% (v/v) TFA followed by cleanup using a Waters HLB plate and sequential elution with 50/50 (v/v) H2O/MeCN with 0.1% FA and MeCN/0.1% FA. After nitrogen drying and lyophilization, samples were reconstituted in 100 microliters of 0.1% FA, a study pool quality control (SPQC) was made by mixing equal volumes of all samples, and 20 microliters of each sample and replicates of the SPQC sample were loaded onto EvoTip Pure tips using manufacturer's protocol. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) used an Evosep One LC with 60 samples per day with a Pepsep 8 cm x 150 micron column. The LC was interfaced to a ThermoFisher Fusion Lumos MS via a FAIMS Pro device with a single CV of -40 V. The precursor scan used a 30,000 resolution mass range from 375-1500 m/z, automatic gain control (AGC) of 4E5 and 54 ms max injection time (maxIT), and ion trap data-dependent tandem mass spectrometry (DDA-MS/MS) used a rapid scan rate, AGC of 1E4 and 45 ms maxIT, an isolation window of 1.2 m/z and normalized higher-energy collisional dissociation of 35%. Cycle time was 1 s. Data was analyzed in FragPipe 20.1 using a 20 ppm precursor and 0.8 product ion tolerance, N-terminal semitryptic specificity and variable proline hydroxylation. A database containing COL1A1/COL1A2 sequences of cow, water buffalo, camel and pig, as well as protein sequences of mollusca, cnidaria and malacostraca, common contaminants and an equal number of reverse sequences (4,936 total entries). Data was annotated at a 1% FDR and visualized using Scaffold 5 (Proteome Software). Per sample spectral counts were exported from Scaffold (Supplemental Data). |
