| Title | Proteomic Profiling of Lipofuscin-Stratified Microglia from Aged Grn Knockout Mice |
| Description | By profiling transcriptomic signatures in aged Grn KO mouse brains, we identified a subpopulation of microglia with high expression of GPNMB and marked lipofuscinosis. To further characterize this population, we isolated lipofuscin "high" and "low" microglia (CD11b+/CD45+) by FACS from the brains of aged (20-month-old) WT and Grn KO mice (n = 4-6/group) and investigated the proteome. Proteomics confirmed an increase in Grn KO specific markers identified by transcriptomics, such as GPNMB. Additionally, proteins belonging to the coordinated lysosomal expression and regulation (CLEAR) network were upregulated, suggesting lysosomal dysfunction in these cells. Finally, myelin-specific proteins were elevated in GrnKO microglia, and some were further elevated in lipofuscin "high" cells, consistent with previous reports of myelin debris accumulating in Grn KO mouse and GRN-FTD patient brain. |
