TitleProteomic profiling of obesity-associated metabolic and immune changes in Göttingen minipigs
DescriptionObesity is a well-established risk factor for increased severity following influenza A virus (IAV) infection, likely through meta-inflammation and immune dysregulation. The mechanisms by which obesity alters antiviral and inflammatory responses remain poorly understood. Using a diet-induced obesity model in Göttingen minipigs, we compared immune and inflammatory responses between obese and lean animals before and after IAV infection. While uninfected pigs showed no clear obesity-associated immune or histological changes in the respiratory tract, obese pigs exhibited reduced viral clearance, altered antiviral gene expression, and increased cellular infiltration post-infection, alongside elevated C-reactive protein levels. This indicates that innate antiviral immunity is affected post-infection in obese animals, potentially contributing to more severe influenza outcomes. The proteomics dataset included here, however, reflects only the obese versus lean state prior to infection, providing insights into baseline metabolic and inflammatory differences. This model demonstrates high translational value for studying obesity-associated immune modulation.
