| Title | rhTPO promotes liver regeneration in rats with acute liver failure |
| Description | This project aims to investigate the effect of recombinant human thrombopoietin (rhTPO) on liver regeneration in rats with acute liver failure (ALF) induced by D-galactosamine (D-GalN). Sixty-six rats were divided into a control group (normal saline injection) and a TPO group (rhTPO injection). After 5 consecutive days of treatment, the ALF model was established. The survival status of the two groups was observed. Platelet count (PLT), liver function indicators (ALT, TBil, etc.), hepatocyte growth factor (HGF), and liver regeneration-related indicators (Ki67, BrdU) were detected at different time points, and transcriptomic and proteomic analyses were performed on liver tissues. The results showed that compared with the control group, the TPO group had significantly increased levels of PLT, serum TPO and HGF, alleviated liver tissue necrosis, increased liver weight index, decreased levels of ALT and TBil, and enhanced liver regeneration capacity. Combined transcriptomic and proteomic analyses revealed that the expression of genes related to cell proliferation pathways such as Mapk1 and Map2k1 was upregulated, while the expression of genes related to inflammatory pathways was downregulated. rhTPO can promote the recovery of liver function, enhance liver regeneration and alleviate liver injury in ALF rats by increasing PLT, inducing cell proliferation and inhibiting inflammation. |
