| Title | Comprehensive prediction and charting of the crustacean neuropeptidomes for Callinectes sapidus and Cancer borealis |
| Description | Background: Neuropeptides are essential regulators of physiological and behavioral processes, yet their discovery in many non-traditional model organisms remains limited by sparse genomic annotation and incomplete sequence information. Here, we present a comprehensive, genome-enabled prediction and validation of the neuropeptidomes of Callinectes sapidus and Cancer borealis, two widely used crustacean model species whose genomes have recently become available. Results: We constructed a curated, cross-species neuropeptide database and aligned known precursors to each genome using tBLASTn, followed by signal peptide detection and peptide prediction. This workflow yielded 14,569 putative peptides in C. borealis and 8,892 in C. sapidus, including numerous sequences exhibiting hallmark features of mature neuropeptides. Integration of mass spectrometry data provided experimental evidence for many predicted peptides and revealed notable isoform diversity, including the first identification of natalisin peptides in C. borealis. Additional findings included novel allatostatin B and allatostatin C isoforms, an insulin-like peptide B-chain-like isoform in C. sapidus, and the discovery that a single precursor can encode two distinct neuropeptide families, an uncommon genomic architecture with potential functional implications. Finally, biological activity evaluation demonstrated robust peptide identification across tissues and revealed feeding-associated regulation in the thoracic ganglion, brain, and commissural ganglion. Several newly identified peptides displayed divergent expression patterns between the pericardial organs and thoracic ganglion, suggesting tissue-specific signaling roles. Conclusion: Together, this work provides the most extensive neuropeptide prediction and validation effort to date in crustaceans, expands the known repertoire of peptide families and isoforms, and establishes a powerful integrative framework for neuropeptide discovery in newly sequenced organisms. |
