TitleAntifungal mechanism of Galla chinensis against Candida albicans by disrupting extracellular vesicles-mediated trafficking
DescriptionTargeting extracellular vesicles (EVs) of Candida albicans as the main pathogen of oral candidiasis, has been proved to be a promising strategy to reduce its drug resistance and virulence. However, the effect mechanism of traditional Chinese medicine Galla chinensis on the fungi remains unclear. Our investigation indicated that aqueous extract of G. chinensis (GCE) exhibited powerful antifungal activities on cell growth and viability, and significantly impaired its ultrastructure, biofilm formation and morphogenesis of C. albicans. Besides, GCE markedly decreased EVs particle concentration by 94% and disrupted the assembly of approximately two-thirds of cargo proteins involved in the pathways of translation and energy metabolism. It significantly downregulated the transcription of genes associated with biofilm formation, hyphal morphogenesis, and virulence, such as Tec1, Bcr1, Hwp1, Cht3, Tos1, Sap6, and Als1. Its effect mechanism is to disrupt EVs-mediated trafficking of cargo proteins, and impair the multi-targets or biological processes including cell growth, and hyphal morphogenesis and biofilm formation of C. albicans and relieved drug resistance. Therefore, GCE is a promising antifungal drug or a vesicle inhibitor for C. albicans. This research makes obvious contribution to a profound understanding of therapeutic mechanism of natural medicine G. chinensis on oral candidiasis.
