| Title | A repressor-activator bridging protein drives transcription factor binding to HDAC/Rpd3L |
| Description | Phenotypic characterization of members of Rpd3L demonstrate the importance of peripheral components including transcriptional repression and activation. To characterize these components, endogenous yeast Rpd3L complex was purified using three baits: Rxt3Tap, Sap30Tap, and Sin3Tap. Cryo-EM structures were acquired on each bait. Taking the Sap30Tap purification, enrichment and crosslinking analysis was performed using mass spectrometry. Integrating this data allowed for an integrated structural model to expand the structure from the cryo-EM core of Rpd3L to a complete model of Rpd3L including its incorporation of Ash1 and Cti6. Further integration of fragment-fragment interaction mapping and crystal structures at the interface of Sin3 PAH2 and Ume6 SID allowed for the incorporation of Ume6. A mutagenesis screen further verified specific residues that are essential for the interaction of Sin3 to Ume6 and Sin3 to Sap30. |
