| Description | This dataset contains LC-MS/MS data acquired to evaluate MR-SP2, a microreactor-based, one-pot sample preparation workflow designed for spatial proteomics of FFPE tissue after laser capture microdissection (LCM). FFPE murine kidney tissue was microdissected using an upward pressure-catapulting LCM system, and defined tissue microregions of 50,000 um^3 (~22 cells), 12,500 um^3 (~5-6 cells), and 3,125 um^3 (~1-2 cells) were isolated. Each LCM-captured specimen was processed either with the MR-SP2 workflow or with a conventional tube-based protocol. The dataset includes raw LC-MS/MS files, sample metadata, and information on specimen size, preparation method, and capture success. The data document improved performance of MR-SP2 across all input levels, including higher capture success rates and increased protein identification depth, especially for few-cell samples. This dataset supports benchmarking of microreacto-based sample preparation for low-input FFPE spatial proteomics and provides reference data for comparing microreactor versus tube-based processing strategies. |
