TitleProteoform-Resolved Phosphorylation Dynamics in Kinase Complexes by Hybrid Precision Mass Spectrometry
DescriptionOur hybrid precision MS strategy integrates three complementary MS approaches to comprehensively characterize phosphorylation dynamics. (i) Intact protein reverse phase liquid chromatography mass spectrometry (RPLC-MS) quantifies proteoform changes over time , (ii) bottom-up MS resolves site-specific phosphorylation kinetics, and (iii) top-down fragmentation using ultra high-resolution MS definitively identifies proteoform architectures through sequencing. This integrated approach addresses individual method limitations by combining intact mass, sequence information, and temporal dynamics in a single workflow .
