TitleIdentification of complex core-fucosylated glycans on proteins associated with metastatic progression in breast cancer
DescriptionEvidence suggests that cancer cells, including breast cancer, have perturbed metabolisms and modified the use of sugars leading to unique expression patterns of N-glycans. In particular, core-fucosylation, a fucose residue added to the core N-glycan structural sugar ,is a major N-glycan modification that is added to proteins and it is significantly upregulated in breast cancer. Analysis of human tumor microarray (TMA) samples by imaging mass spectrometry (iMS) identified core-fucosylated N-glycans (ie. the product of having high FUT8) that are strongly associated with breast cancer metastasis8. Inhibition of core-fucosylation in metastatic breast cancer cells inhibits cell migration, invasion, and metastatic signaling. ere, we report methods to use the the lectin rAALN224Q, which was previously characterized to bind specifically to complex core-fucosylated N-glycans, and show that we can successfully use rAALN224Q (heretofore referred to as N244Q) to enrich proteins and identify glycopeptides using high resolution LC-MS/MS . Initial N244Q enrichment and glycoproteomic analysis using 4T1 cells in the presence or absence of 2FF resulted in identification of core-fucosylated proteins with significant relationship to metastasis related pathways. Further analysis identifies complex core-fucosylated N-glycans on specific proteins including LRP1, THBS1, and cMet. Blockade of fucosylation alters the interactome of these core-fucosylated proteins leading to changes in their signaling capacity.
