Title	Comprehensive Proteomic Analysis of Colorectal Cancer Liver Metastasis
Description	Colorectal cancer (CRC) liver metastasis (CCLM) is a major driver of CRC-related mortality, yet its molecular mechanisms remain incompletely understood. This study performed comprehensive proteomic analysis using 38 tissue samples: 10 benign adenomas (control, CONT), 10 primary CRC (PCRC), 10 liver metastases (HM) for data-independent acquisition (DIA) profiling, and 8 additional samples (4 PCRC, 4 HM) for parallel reaction monitoring (PRM) validation. Principal component and clustering analyses revealed distinct proteomic signatures among groups, with partial overlap between PCRC and HM indicating shared tumorigenic profiles. Pairwise comparisons identified 1,719 up/168 downregulated proteins in PCRC vs. CONT, 1,366 up/148 down in HM vs. CONT, but only 89 differentially expressed proteins in HM vs. PCRC, suggesting liver metastasis involves targeted regulation of key proteins rather than widespread reprogramming. Functional enrichment highlighted pathways in extracellular matrix remodeling, immune evasion, and coagulation as critical for metastasis. Three biomarkers were prioritized: HDHD3 (AUC = 0.830, upregulated in HM), OLFM4 (AUC = 0.760), and ZNF34 (AUC = 0.860, overexpressed in PCRC), implicating roles in metastatic progression. These findings provide insights into CCLM mechanisms and potential targets for diagnosis and therapy.
