TitleImportin 7 - HIV-1 integrase LC-MS/MS
DescriptionThis project investigates the interaction between Importin 7 (Imp7), a nuclear transport receptor, and HIV-1 Integrase (IN), a key viral enzyme required for integration of the viral DNa to the host genome. Although Imp7 has been implicated in facilitating nuclear entry of the HIV-1 IN, the molecular details of its interaction with IN remain unclear. To characterize the physical interaction between Imp7 and IN, we applied chemical cross-linking mass spectrometry (XL-MS) using BS3 as a cross-linker. The complex was expressed, purified, and cross-linked under defined conditions, followed by tryptic digestion and LC-MS/MS analysis on an Orbitrap instrument. Five technical replicates of the same IMP7–IN complex were analyzed to ensure data robustness and reproducibility. Identified inter- and intra-protein cross-links reveal putative contact sites between Imp7 and IN, providing insight into the structural basis of their interaction. This dataset supports our broader aim of understanding host–virus interactions at the molecular level and contributes to the structural biology of nuclear import mechanisms in the context of HIV infection. All raw data files, cross-link identifications, and metadata are provided in this submission.
