Description | Background: Stem cell transplantation has brought a new method for the treatment of ischemic heart disease, but its slow proliferation, weakened migration ability and decreased differentiation potential in situ are urgent problems to be solved before clinical application. Promoting blood circulation and removing blood stasis is one of the basic methods for treating ischemic heart disease with traditional Chinese medicine, which has achieved good clinical effect. Ligusticum Chuanxiong (Chuanxiong) and Radix Paeonia (Chishao) is a commonly used drug pair for promoting blood circulation and removing blood stasis. The main objective of this study was to determine whether the combination of Chuanxiong- Chishao with stem cell transplantation could synergically promote angiogenesis in ischemic myocardium. Methods: A myocardial infarction (MI) model was established by ligation of branch of the left anterior descending coronary artery in SD rats. Rats were randomly divided into the sham operation group, model group, MSCs group and Chuanxiong-Chishao combined MSCs (CC-MSCs) group. MSCs were injected into the myocardium immediately after ligation, and the herb pair was administered for 28 days. The left ventricular function of the rats was measured by ultrasound. Myocardial infarction size and histopathological changes were observed by Masson staining, and CD31 levels were detected by immunofluorescence. Samples from the model and CC-MSC groups were analyzed by label-free quantitative proteome assay, and the target proteins were verified by Western blotting. Results: Compared with MSCs alone, CC-MSCs improved LVEF, FS and CO values, reduced the myocardial infarction area and increased the number of new vessels in MI rats. Proteomic results showed that 154 differentially expressed proteins were obtained after CC-MSCs were compared with the model group. Protein enrichment analysis showed significant differences in "positive regulation of angiogenesis" and "positive regulation of vasculature development". Among differentially expressed proteins, expression levels of CDC42, NRP1 and Amot were significantly changed. Western blot results showed CDC42, NRP1 and Amot were consistent with the proteomics results. Conclusions: Chuanxiong-Chishao combined with MSCs has a synergistic effect on promoting angiogenesis of ischemic myocardium, and the CDC42-mediated signal axis may be the key mechanism. |
