Dataset Summary
| Title | Response Marker Enrichment Analysis informs the impact of pharmacological and genetic perturbagens on cancer cells |
| Description | The project focuses on the development and validation of our novel enrichment technique, Response Marker Enrichment Analysis (ReMEA). As part of this validation, we present two datasets consisting of MS/MS analysis of drug treated acute myeloid leukaemia (AML) cell lines. These are as follows: Firstly, we present a total proteomics dataset, whereby the HEL and P31-Fuj cell lines were treated with lysine-specific demethylase 1 (LSD1) inhibitors for 5-days. Here, cells were exposed to two 1µM treatments of either GSK2879552 or iadademstat. This data was then analysed to infer PI3Ki sensitivity as described in the publication. Secondly, we present a phosphoproteomics dataset, consisting of chemotherapy treated cell lines. The cell lines used were the P31-Fuj, MV4-11 and HL-60 cell lines. The chemotherapy treatments consisted of cytarabine, daunorubicin and a combination of the two for either 2, 6, or 24 hours. Cytarabine was used at 2µM, daunorubicin 1µM and co-treatment doses were also 2µM and 1µM for cytarabine and daunorubicin, respectively. This data was then analysed and used to construct our compound activity marker signatures, with which we stratify AML patients using published phosphoproteomics data (as described in the publication). |
