| Description | In the present study, lysozyme was analysed in the Venetin-1 protein-polysaccharide complex obtained from the coelomic fluid of the earthworm Dendrobaena veneta and exhibiting anticancer, antifungal, and immunostimulatory activity. The analysis of fractions with different molecular weights revealed the highest concentration of lysozyme recognised by antibodies directed against human lysozyme in the fraction containing compounds with a mass above 100 kDa and in the complex obtained using dialysis bags with a cut-off point of 6-8 kDa. The activity of lysozyme in this preparation was 1181.43 U/mg. FTIR analyses confirmed the validity of using a lower cut-off point and showed greater similarity of the spectrum of the complex obtained with this method to hen egg white lysozyme (EWL). Raman analysis showed significant similarity of the protein part of Venetin-1 and EWL. Cryo-EM studies revealed the structure of the tested nanoparticle with similarity to lysozyme and lysenin. Immunoblotting using antibodies directed against human lysozyme showed proteins with a mass of 11, 33, 44, 88, and 132 kDa recognised in Venetin-1. These observations suggest the presence of polymeric forms of lysozyme in the tested complex. Zeta potential analysis revealed properties of the nanoparticle that predispose it to be used in medicine. |
