| Title | Premature mesocarp vitality loss in Niagara Rosada occurs independently of cell death |
| Description | The grape berry mesocarp undergoes physiological and biochemical changes during ripening, including mesocarp cell death (MCD) and hydraulic isolation. We hypothesized that MCD does not occur in the Niagara Rosada (NR) variety but rather, a down-regulation of respiratory enzymes during the early loss of mesocarp cell viability while preserving membrane integrity. Hence, we analysed three developmental stages using label-free proteomics, enzymatic respiratory activity assays and outer mesocarp imaging. Cell wall-modifying proteins accumulated differentially throughout ripening, while cytoplasmic membranes remained intact. Additionally, the reduction in the mitochondria cristae density coincided with decreased malate dehydrogenase and succinate dehydrogenase activities. Proteomics analysis revealed 956 differentially accumulated proteins, with most down-regulated at ripening. However, the enzymes aldolase, glyceraldehyde-3-phosphate dehydrogenase, phosphoglycerate kinase, enolase and malate dehydrogenase were among the most abundant at ripening or contributed significantly to data variance, suggesting the maintenance of respiratory activity despite mesocarp physiological changes. We also hypothesized that gluconeogenesis originated from malate occurs in mesocarp, and that sucrose futile cycles may play a crucial role in carbohydrate storing and unloading. Therefore, we suggest that the premature loss of berry mesocarp vitality in NR is not linked to cell death. |
