| Title | Polystyrene Microplastics effects on human gingival fibroblasts |
| Description | Human gingival fibroblast cells were treated with 50 micro grams per mL of 1 micro meter polystyrene microplastics, and the control (with only the medium) after 48, and 72 hours the cells were collected to obtain a pellet. Total cell extracts were obtained by treating cells with RIPA buffer 0.1 SDS plus protease and phosphatase inhibitors then incubated on ice. After centrifuging at12000 rpm for 20min at 4C. The Pierce BCA Protein Assay Kit to determine the protein concentration. A volume corresponding to 25 micro grams of proteins was loaded into a Nanosep 10-kDa-cutoff filter and digested according to our routine protocol. Each digested protein sample was analyzed in technical triplicate. Raw data were processed using PEAKS studio using the correct precursor only option. Spectra were matched against the UniProt SwissProt database restricted to Homo sapiens taxonomy, to which a list of common contaminants was appended. |
