Description | Endoplasmic reticulum is a considerable network of intracellular network. Major functions of the ER are a protein folding, proteosynthesis, and post-transcriptional modifications, lipid anabolism and sorting of the protein. ER isolation is a very challenging because ER biochemical properties are mostly like the other cellular endomembrane. Most previously published protocols for endoplasmic reticulum isolation are based on the density gradient ultracentrifugation. Although it is a sub optional resolving power. In this comprehensive study, we mainly focused on the endoplasmic reticulum of developing chickpeas to gain an overview of proteomics analysis. Easy-nlc LC-MS/MS technique of proteomics was utilized as the main balancing method to finding large-scale identifications of the endoplasmic reticulum proteins. ER protein identification from the Easy-nlc LC-MS/MS proteomics techinque was carried out using the Amanda search engine with the Uniprot Cicer Arietinum V2.5 database. It was the objective of this analysis to exhibit and exploit the capabilities of Easy-nlc LC-MS/MS in proteomics characterization of the tissue of chickpea leaf, with a particulular focus on the endoplasmic reticulum. Using this techinque and digestion by the trypsin, we identified over 2193 proteins across total endoplasmic reticulum samples. Here, we optimised the endoplasmic reticulum isolation protocol from chickpea tissue. Combined with the enzymatic analysis and proteomics analysis, this data will facilitate our better understanding towards the ER proteomics study in chickpea plants. |
