TitleCX3CR1 modulates migration of resident microglia towards brain injury
DescriptionMicroglia are the innate immune cells of the brain. They are highly motile cells that screen the surrounding brain parenchyma on a minute timescale. In an ATP-dependent manner, microglia extend their processes towards an injury in the central nervous system (CNS). Microglia migrate towards ischemic sites within hours in vivo. However, whether the fractalkine receptor (CX3CR1) influences microglial migration remains unknown. Label free proteomic profiling pinpointed elevated motor dysfunction and differential changes in Rho GTPase signaling activity that indicated a dysregulated actin cytoskeleton signaling in Cx3cr1-deficient cortex. To investigate migration properties of microglia, we carried out 4-hour interval two-photon in vivo imaging for 72 hours after a laser lesion in the cortex. Cx3cr1-deficient mice showed enhanced migration towards the lesion. Not only cell body migration, but also length and velocity of microglial fine processes extending towards the lesion were increased in Cx3cr1-deficient microglia. Migration and number of migrating microglia remained unchanged in Ccr2-deficient mice, indicating that monocyte-derived macrophages/microglia did not contribute to migration and proliferation around the injury. These results demonstrate microglia migration towards CNS injury and suggest a role for CX3CR1 in modulating this process. Manipulating microglia migration may become an important target for the treatment of CNS-injury.
