Description | PTPRD is a presynaptic adhesion molecule implicated in various neurological, psychiatric, and neurodevelopmental disorders, but the underlying mechanisms remain unclear. PTPRD trans synaptically interacts with multiple postsynaptic adhesion molecules, thorough the regulation by its extracellular alternatively spliced mini exons (meA and meB). While PTPRD meA regulates specific hippocampal excitatory synapses, PTPRD meB has not been studied in vivo. Here we found that homozygous PTPRD meB mutant (Ptprd meB homo del) mice show early postnatal lethality and that heterozygous Ptprd meB hetero del mice show behavioral and synaptic abnormalities, which are not observed in PTPRD meA mutant mice. In Ptprd-meB hetero del mice, excitatory synapses in dentate gyrus granule cells (DG GCs) show decreased density and transmission, whereas PTPRD meA mutant mice show normal excitatory synaptic transmission. Proteomic analyses indicate decreased postsynaptic levels of IL1RAP, a known trans synaptic partner of PTPRD meB. Accordingly, IL1RAP mutant mice show similarly decreased excitatory synaptic transmission in DG GCs. Therefore, PTPRD meB is more important than PTPRD meA for postnatal survival, excitatory synaptic function, and mouse behaviors, and IL1RAP mediates PTPRD meB dependent excitatory synaptic regulation. |
