TitleCompound-centric chemical proteomics to screen fructose binding proteins
DescriptionTo identify protein targets of fructose, we performed a compound-centric chemical proteomics (CCCP) experiment on podocytes with P-fructose. Briefly, podocytes were incubated with P-fructose, and the cellular proteins that directly bind to fructose were covalently crosslinked to P-fructose by UV irradiation. After orthogonal conjugation was achieved with azide-biotin via click chemistry and affinity purification was performed, the P-fructose-enriched proteins were analyzed by liquid chromatography coupled with tandem mass spectrometry (LC‒MS/MS).
