To explore the specific alteration in host epithelial EVs induced by glucocorticoid, we performed liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based proteomic analysis on EVs derived from HOKs pretreated with glucocorticoid or not. The results showed a total of 389 differentially expressed proteins . Among the top 10 upregulated proteins, most were extracellular matrix (ECM) components ). Enrichment analysis of the relevant proteins revealed that the upregulated GO terms were centered on collagen binding, cell adhesion, and extracellular matrix component aggregation. Meanwhile, the associated biological processes included epithelial cell proliferation and differentiation, fibrin synthesis, regulation of epithelial healing, and barrier repair . The volcano plot revealed that, among the top ten most significantly upregulated proteins, two ECM proteins Lumican and Biglycan are closely related to barrier repair.
