Chow diet‑fed (Chow) and high-fat diet-fed (HFD) mice were maintained under ad libitum feeding (Fed) or 24 h fasting (Fasted). Pancreases were harvested (n = 4 per group), ground in liquid nitrogen, and lysed in Buffer A (50 mM Tris-HCl pH7.4, 150 mM NaCl, 1 mM EDTA, 1% TritonX-100, 4M Urea). Lysates were centrifuged at 4oC for 10 min at 12, 000 rpm, and the resulting supernatants were collected. Protein concentrations were determined using the BCA assay prior to mass‑spectrometry‑based proteomic analysis.
