HDX-MS experiments were performed as described in detail in Białobrzewski M.K., Cieplak-Rotowska M.K., et al (https://www.biorxiv.org/content/10.64898/2026.07.16.738963v1) on human CNOT1 (residues 800-999) in the presence of peptides corresponding to GW182 CIM1 (QSRLPQWTHP), N-CIM1 (DPSQSQSRLPQWTHPN), the GW182 SD10 fragment (TNRC6C residues 1260-1620), or the TTP peptide (APRRLPIFNRISVSE). The interaction was additionally quantified by performing HDX-MS titrations of CNOT1(800-999) with increasing concentrations of the N-CIM1 peptide (0, 1.4, 3.5, 14, 70, 140, 280, and 700 µM).
