The interferon inducible isoform of human NCOA7 (NCOA7-AS, for Alternative Start) is a TLDc protein that has been shown to be antiviral against influenza A virus and to interact with cytoplasmic subunits of the V-ATPase. Upon its ectopic expression, there is a decrease in endolysosomal pH that seems detrimental to the virus. In order to gain further light into NCOA7-AS mechanism of action, we have performed co-immunoprecipitation experiments (using triple flag tagged NCOA7-AS in U87-MG cells as well untagged NCOA7-AS as a control) and mass spectrometry analysis. Of note, U87-MG cells were used as they are easy to transduce with lentiviral vectors and express high levels of NCOA7-AS.
