This study investigates how differentiated white adipocytes adapt to cooler temperatures by remodeling their acetyl-lysine–modified proteome. Differentiated adipocytes were cultured at 37°C or 31°C for 12 days to induce temperature-dependent physiological and biochemical changes. To profile acetylation dynamics on a global scale, proteins were extracted, digested with trypsin, and subjected to immunoprecipitation (IP) of acetyl-lysine–modified peptides. The resulting enriched peptides were analyzed by mass spectrometry to identify and quantify acetylation sites, enabling comparison of acetylation patterns between cells cultured at 37°C and 31°C.
