The cells were taken out of the incubator, washed three times with pre-cold PBS, then scraped into an EP tube using a clean cell scraper, centrifuged at 4 degrees for 5 minutes to remove the upper PBS, and then rapidly frozen in a -80 degree freezer for testing.
