This project develops the photocatalytic lysosome-tracking proteome mapping (PLT-PM) method, which uses LysoTracker DND-26 as both a lysosomal tracker and visible-light photocatalyst. The method enables one-step, high-resolution, and low-disruption profiling of the lysosomal proteome without requiring genetic modification or custom dye synthesis. It was applied alongside LC-MS/MS to analyze lysosomal proteome changes during rapamycin and amino acid starvation-induced autophagy, offering a biocompatible and accessible platform to advance lysosome-related research.
