We prepared nuclear extracts from three types of HeLa cells: those expressing ICE1::AFF4, ICE1 alone, and untransduced control cells. Using V5-tag immunoprecipitation (IP), we aimed to compare the proteins associated with ICE1 and ICE1::AFF4. Our hypothesis is that the ICE1::AFF4 fusion protein will pull down more components of the Super Elongation Complex (SEC) than ICE1 alone, indicating enhanced interaction due to the presence of AFF4. This would suggest that AFF4 promotes SEC recruitment when fused to ICE1, potentially modulating transcriptional elongation.
