TitleReceptor kinase pathway signal tuning through a non-transcriptional incoherent feedforward loop
DescriptionThis project investigated an auto-regulatory feedback in a plant receptor kinase pathway: The signaling of the endogenous CLAVATA3/EMBRYO SURROUNDING REGION 45 (CLE45) peptide via the receptor kinase BARELY ANY MERISTEM 3 (BAM3) in Arabidopsis (Arabidopsis thaliana) vascular development. Downstream, the plant-specific MEMBRANE-ASSOCIATED KINASE REGULATOR (MAKR) family protein MAKR5 conveys CLE45-BAM3 signaling, which is amplified by CLE45-BAM3-triggered enhancement of MAKR5 production through the stimulation of MAKR5 mRNA translation. At the same time, this signal amplification is dampened through CLE45-stimulated MAKR5 phosphorylation, which inactivates MAKR5, enhances its turnover and impinges on MAKR5 mRNA levels. In summary, the results suggest a non-transcriptional incoherent feedforward loop in which CLE45-BAM3 signaling is amplified via translation of MAKR5 mRNA yet also balanced via phosphorylation of MAKR5 protein.
