Toxoplasma gondii is a ubiquitous eukaryotic pathogen. Apicomplexan parasites-the family to which T. gondii belongs-possess a divergent ortholog of mammalian PDK1. We have previously shown that this store potentiating/activating regulatory kinase (SPARK) is essential for T. gondii motility, invasion, and egress. Here, we conditionally depleted SPARK in live parasites for 24 hours to determine the impact of SPARK function on the parasite proteome.
