TitleSphingosine Kinase-2 Inhibition Increases Acetyl-CoA Carboxylase Activity
DescriptionThe presence of immunosuppressive cells, including myeloid-derived suppressor cells (MDSCs) and tumor-associated macrophages (TAMs), within the tumor microenvironment (TME) represents a significant challenge in adoptive T cell therapy. Consequently, strategies designed to improve anti-tumor T cell function in vivo by mitigating this immunosuppression are essential. Here in, we demonstrate that inhibiting sphingosine kinase-2 (SphK2) mediated sphingosine-1-phosphate (S1P) generation in myeloid-derived suppressor cells (MDSCs) significantly reduces their suppressive potential and promotes their antigen presenting capacity. An S1P pull-down assay, followed by mass spectrometry-based proteomics analysis, demonstrated that S1P interacts with multiple proteins essential for metabolic processes. Both isoforms of Acetyl CoA Carboxylase (ACC1 and ACC2), which are the rate-limiting enzymes in fatty acid metabolism, exhibit binding to S1P.
