| Description | Our hybrid precision MS strategy integrates three complementary MS approaches to comprehensively characterize phosphorylation dynamics. (i) Intact protein reverse phase liquid chromatography mass spectrometry (RPLC-MS) quantifies proteoform changes over time , (ii) bottom-up MS resolves site-specific phosphorylation kinetics, and (iii) top-down fragmentation using ultra high-resolution MS definitively identifies proteoform architectures through sequencing. This integrated approach addresses individual method limitations by combining intact mass, sequence information, and temporal dynamics in a single workflow . |
