Description | Photoaffinity probe-based protein labeling of microbial isolate Acinetobacter pittii SO1 (NCBITAXON:48296), isolated from the sorghum rhizosphere that can utilize sorgoleone as a sole carbon source in order to identify proteins that bind sorgoleone. Photoaffinity labeling of A. pittii lysates using the SoDA-PAL probe approach may be extended in the future to proteomic profiling of complex rhizosphere microbiomes to discover genes that can be leveraged to promote beneficial plant-microbe interactions (grown on both acetate and purified sorgoleone). Eluted peptides from the C18 column were analyzed using a Q-Exactive Plus Orbitrap for high resolution MS and high-energy collision-induced dissociation tandem MS by electrospray ionization for subsequent quantitative proteomic analysis. |
