Description | Scope: 4-Methylcatechol (4MC) is a polyphenol with origin in processed food. Protein-polyphenol adducts are formed upon covalent bonding between oxidized polyphenols and proteins in food. The present study investigated in vitro gastric and intestinal digestion of a milk protein, beta-lactoglobulin (beta-LG), covalently modified at its nucleophilic amino acids by 4-methylbenzoquinone (4MBQ), the oxidized form of 4MC. Present results characterize 4MBQ-induced covalent modifications on beta-LG and their effect on protein digestibility. Methods and results: The beta-LG-4MBQ adduct (beta-LQ) was prepared at pH 8 by reacting beta-LG and 4MBQ in equimolar ratio for 10 min at 22 degrees Celcius. Significant thiol and amine loss was found in beta-LQ compared to beta-LG. 4MBQ-induced modifications were identified on Cys, Lys, Arg, His and Trp in beta-LQ by LC-MS/MS after tryptic hydrolysis. The concentration of Cys-4MC adduct was determined after total acidic hydrolysis of beta-LQ. No significant differences between beta-LG and beta-LQ on in vitro digestibility were observed by assessment with SDS-PAGE, degree of hydrolysis and LC-MS/MS peptide intensities. Nine of the in vitro digested peptides from beta-LG and beta-LQ were identified as bioactive from a database search. Cys-4MC adduct was found to be bioaccessible after in vitro digestion. |
