TitleGIGYF2 regulates telomere length and cellular senescence by restricting telomerase access
DescriptionThis dataset presents the proximity-dependent biotinylation (PhastID) proteomic profiling of GIGYF2, a newly identified negative regulator of telomerase. Human cells expressing BirA-fused GIGYF2 were labeled with 50 μM biotin for 4 hours. Biotinylated protein complexes were captured using streptavidin magnetic beads, followed by on-bead reduction, alkylation, and tryptic digestion. The resultant peptides were analyzed by LC-MS/MS on an Orbitrap Fusion Lumos instrument. Data processing was performed using MaxQuant (v2.1.0.0) against the UniProt human database. Protein abundance was normalized by calculating Fraction of Target (FOT) values based on iBAQ intensities. This interactome analysis reveals GIGYF2's association with the CCR4-NOT complex and the shelterin components TPP1/POT1, supporting its role in post-transcriptional regulation and telomere homeostasis. This resource contributes to the mechanistic understanding of telomere biology and potential therapeutic targeting in aging.
