TitleCharacterization of the proteome of P. aeruginosa under different combinations of cAMP and c-di-GMP levels
DescriptionThe aim of this experiment was to assess proteomic changes in P. aeruginosa PAO1 under different combinations of intracellular cAMP and c-di-GMP levels. Intracellular c-di-GMP levels were manipulated by expressing the P. aeruginosa phosphodiesterase PA5295 (LG) or the diguanylate cyclase WspR19 (HG) from pBBR5-MCS. The empty plasmid served as the unaltered c-di-GMP condition (FG). Intracellular cAMP signaling was reduced by deletion of the adenylate cyclase genes cyaA and cyaB (LA), or increased by deletion of the cAMP phosphodiesterase gene cpdA (HA). The parental background served as the unaltered cAMP control (FA). Each cAMP genotype was combined with each c-di-GMP genotype, yielding a nine-state signaling toolbox. Cultures were grown overnight at 37 C with agitation in MOPS based minimal medium supplemented with either glucose and NH4+ (-G-N) or glucose and choline (-G-C).
