TitleProteomic profiling of THP1 wild‑type and ARG2‑knockout human monocytes under normoxic and hypoxic conditions
DescriptionArginase 2 (ARG2) is a mitochondrial enzyme implicated in metabolic and inflammatory regulation in human monocytes. To investigate the proteomic consequences of ARG2 loss and its interaction with hypoxic stress, we performed a quantitative LC‑MS/MS analysis of THP1 wild-type (THP1WT) and ARG2‑knockout (THP1ARG2‑/‑) cells. Cells were synchronized by serum starvation and subsequently exposed for 48 hours to either normoxia (21% oxygen) or hypoxia (1% oxygen), which generates four experimental groups: THP1WT-normoxia, THP1WT‑hypoxia, THP1ARG2‑/‑‑normoxia, and THP1ARG2‑/‑‑hypoxia. Each condition was analysed in biological triplicate (12 samples total). The dataset provides a comprehensive overview of baseline proteomic differences between THP1WT and THP1ARG2‑/‑ monocytes, as well as their differential responses to hypoxic stimulation, offering a detailed resource on ARG2 related and hypoxia induced molecular changes in THP1 cells.
