| Description | NAD depletion is a widespread bacterial defense strategy against phage infection. In turn, phages have evolved NAD reconstitution pathways (NARPs) to replenish NAD and sustain infection. However, the molecular basis underlying NARP function remains poorly understood. Here, we uncover the structural and mechanistic basis of ADP ribose pyrophosphate synthetase (Adps) and nicotinamide ADPR transferase (Namat) in the NARP1 pathway. Structural snapshots of Adps reveal a dramatic 22 A catalytic loop closure that recognizes the adenine moiety of ADPR and simultaneously drives active-site completion and intermediate stabilization. Adps structures bound to ATP and ADPR analogs support a sequential Bi Bi mechanism in which ATP binding primes subsequent ADPR recognition, followed by ordered AMP and ADPR-PP release. Adps lacks allosteric sites for ADP inhibition, but instead assembles with Namat containing a distinct insertion domain into higher order complexes that facilitate NAD reconstitution. Overall, these findings advance our understanding of NAD synthesis through NARP1 pathway and the bacterial phage arms race |
