TitleMulti-Omics Study of the Therapeutic Targets of Tribulosin D in Pulmonary Fibrosis
DescriptionPulmonary fibrosis (PF) is a progressive and refractory lung disease with limited therapeutic options. Tribulosin D (TED) has demonstrated anti-fibrotic activity; however, its underlying molecular mechanisms remain unclear. In this study, a bleomycin-induced PF mouse model was established to evaluate the therapeutic effects of TED. Histopathological staining and ELISA results showed that TED markedly reduced inflammatory cell infiltration and collagen deposition in lung tissues. To elucidate its mechanism of action, an integrated multi-omics strategy combining transcriptomics, proteomics, metabolomics, bioinformatics, and machine learning analyses was employed. Potential therapeutic targets were identified through differential expression analysis, public database integration, and LASSO, random forest, and SVM-RFE algorithms. The results revealed Scarb and Cntfr as key targets, with Cntfr identified as the core target. Functional analyses indicated that Cntfr was involved in the fibrosis-related JAK–STAT signaling pathway and regulated by specific transcription factors. Metabolomic analysis suggested that TED primarily modulated amino acid and lipid metabolism. Molecular docking further confirmed a stable binding interaction between TED and Cntfr. These findings provide new insights into the anti-fibrotic mechanisms of TED and support its potential therapeutic application in PF.
