TitleCirculating cochlin LCCL domain binds apoptotic and necrotic cells and enhances efferocytosis
DescriptionClearance of dead cells by efferocytosis is a critical process for homeostasis as, in particular, it limits inflammation. Defect in efferocytosis has been associated to autoimmune, neurodegenerative and cardiovascular diseases, as well as to chronic infections. Better understanding of efferocytosis regulation may pave the way to therapeutically target this process. Here, we identified circulating cochlin LCCL domain as a new regulator of efferocytosis. While the protective role of cochlin in the antibacterial response of the host is now well-established, the underneath mechanisms remained unknown. Using cell binding assay for recombinant cochlin LCCL domain, we established its specific tropism for apoptotic and necrotic but not pyroptotic and necroptotic cells, of both immune and non-immune lineage, from murine as well as human origins. By three independent functional assays, we demonstrated that endogenous and exogenous cochlin LCCL domain enhances macrophage efferocytosis in vitro and in vivo in LPS-induced inflammation and Pseudomonas aeruginosa intranasal infection contexts. Our findings provide a new role of cochlin LCCL domain in the regulation of efferocytosis, in addition to its described proinflammatory role, as an immunomodulator for host response and homeostasis.
