TitleLoss of ULK1 Promotes TLR4 Acetylation in Bone Marrow-Derived Macrophages
DescriptionThis study focuses on the regulatory effect of ULK1 (Unc-51-like autophagy activating kinase 1) deficiency on TLR4 (Toll-like receptor 4) acetylation in bone marrow-derived macrophages (BMDMs). To systematically explore the regulatory interplay between ULK1 and TLR4 acetylation, primary BMDMs were isolated from wild-type (WT) and ULK1 knockout (ULK1KO) mice and further induced and differentiated into mature macrophages in vitro. Total cellular proteins were extracted from WT and ULK1-deficient BMDMs, and endogenous TLR4 protein was specifically enriched via immunoprecipitation (IP) using a TLR4-specific antibody. The immunoprecipitated protein samples were subsequently subjected to mass spectrometry (MS) analysis to profile the acetylation modification landscape of TLR4. MS screening results successfully identified distinct acetylation modification sites on TLR4.
