TitleNMNAT2 Loss in Primary Mouse Cortical Neurons at DIV8 and DIV12
DescriptionProteinopathy and metabolic dysfunction are central hallmarks of neurodegenerative disease, yet the mechanisms linking these pathologies remain unclear. Here, we investigate the consequences of neuronal nicotinamide mononucleotide adenylyl transferase 2 (NMNAT2) loss, a key regulator of NADplus metabolism. We show that NMNAT2 deficiency in neurons results in widespread proteome remodeling of pathways involved in protein quality control and transport, mitochondrial function, and cellular metabolism. WT control and KO neurons were plated on 12-well plates at a concentration of 1x106 cells/well. At DIV8 or 12, cells were rinsed 3x with ice-cold PBS. 300ul of PBS was added to each well and cells were detached from the plate using a sterile cell scraper. 2 wells were combined into 1 sample and centrifuged at 300g for 8 minutes. Cell pellets were rinsed once with ice-cold PBS and centrifuged again at 300g for 6 minutes. Samples were flash frozen in liquid nitrogen and stored at -80 C. 2022_04_80: 6 wildtype neuron sample and 6 NMNAT2 KO neuron samples collected at DIV12 2022_07_134: 4 wildtype neuron samples, 4 NMNAT2 KO neuron samples, 4 wildtype neuron samples treated with 1mM NMN, and 4 NMNAT2 KO neuron samples treated with 10mM NMN. All collected at DIV8
