DescriptionDormancy release and growth resumption represent a critical developmental transition in perennial geophytes. Lysine acetylation (LysAc) is a key reversible post-translational modification (PTM) that plays an important role in plant development, whereas the mechanism by which lor-miR168-5p-LoAGO1B and acetylation synergistic regulation in geophytes dormancy release remains elusive. Here, our comparative lysine acetylomic analysis of lily bulbs reveals that extensive protein lysine deacetylation was detected during dormancy release. A total of 14,236 LysAc (Kac) sites on 5,778 proteins were identified in lily bulb, enriched in ribosome biogenesis, the TCA cycle, and ncRNA metabolism pathways. Notably, LoArgonaute 1B (LoAGO1B), a core component of the RNA-induced silencing complex (RISC), contains three dynamically changing Kac sites located within conserved PAZ and PiWi domains during dormancy-growth transition. Simultaneously, lor-miR168-5p negatively regulates LoAGO1B at the post-transcriptional level. ‌The Kac sites of LoAGO1B protein and lor-miR168-5p target regulation contributes to the discordance between the transcriptomic and proteomic variations during growth transition. Acetylome reprogramming is driven by specific lysine deacetylases (KDACs) and acetyltransferases (KATs). Critically, KDAC inhibition greatly altered LoAGO1B expression and Acetyl-CoA activity. Virus-induced gene silencing (VIGS) experiments revealed that LoAGO1B plays a positive role in regulating dormancy release by modulating global LysAc level. Collectively, our findings reveal a novel epigenetic regulatory axis integrating miRNA action and LysAc to the rewiring of geophyte dormancy release.
